Well, the best and almost broken is this unarmed (maybe a knife) high Dex build that utilize Fatal Throw. THere was topic somewhere near.
Are you sure you want to read it?
Underrail system is simple and not overly complicated like in PoE e.g., there is nothing difficult about it, simple but deep and flexible system.

Heavy Assault rifle builds are quite effective.

They will also die a lot less than the unarmed build, due to being so tanky.

It's REAL FUN to try a lot of builds. Many of them are perfectly viable. Just experiment and have fun.  8)

No idea what the best build is, since you can't do everything with one character, and different people will argue that different things are indispensible.

I would say SMG specialists have extremely high damage output due to the fact that they can burst many times per round, and the build synergises well with stealth, dodge, evasion, throwing, traps and lockpicking. However you won't ever be very good at crafting if you invest in all of those skills and will have to rely on stuff you find or buy. Plus you'll have to use light armor and probably also dump constitution, so you'll be a glass cannon for a lot of the game.

Yeah to summarize what people are saying, every build has its own strength.
----------------------------------------------------------------------------------
If you want a build that has no weakness, you won't find one. The strongest builds fail at times too, there's no "One build to rule them all" sort of thing in this game.

For instance, AR builds are extremely tanky and easy to understand and play in easy and normal, but might have issues in Hard mode due to lack of stealth and initiative in combat.
More trap oriented/Stealthy builds show up in Hard mode, although there are plenty of amazing "glass cannon" builds for HM as well.

I recommend Wildan's Sledgehammer Build for a good all around build for Easy-Hard.  http://underrail.com/forums/index.php?topic=1736.0
